Budget hotel chain Travelodge has received planning permission from the mayor for a 188-bedroom hotel close to the entrance of the Olympic Park in Stratford.
The new Travelodge hotel in Stratford will be located at 1-4 Park Lane, E15.
More than 30 jobs will be created, with all entry level roles going to the local long-term unemployed via a partnership with Job Centre Plus.
In February the group announced it will add another 10 sites to its portfolio, which have a combined investment value of £60.7m, as it continues its push into city centres.
The plans, which included the Stratford site, keep the group on track to become the single biggest hotel brand in London by the time of the Olympics, with a total of 45 hotels.
Travelodge chief executive Grant Hearn has also written to the Olympics minister asking her to confirm or deny the rumour that Visit Britain is to receive a budget cut of 20%.
He said: “The Olympics provide us with a once in a generation opportunity to promote London globally yet the government is showing incredible apathy towards maximising the potential.
“Already operating under financial strain it appears Visit Britain is set to be on the receiving end of a 20% budget cut right at the time we should be increasing our exposure to the world.
“More must be done domestically to capitalise on the Games coming to London.
“Where are the banners on the streets or the welcoming signs at Kings Cross and Heathrow?
“Complacency has set in and we must not allow the enormous potential benefits for London’s tourist economy slip through our fingers.”
