The route for the second phase of the new HS2 railway line, linking Birmingham with Leeds, will mean the demolition of 16 newly built homes on the Shimmer estate in Mexborough, the Department for Transport said.
The news comes after the government dropped plans for a stop at the Meadowhall shopping centre in favour of using the existing Sheffield city centre station.
The government also awarded contracts for the initial phase of the project, with £1.4bn of work going to a consortium that includes troubled construction company Carillion.
